How do you get liquid culture? ›

To make liquid culture, mix a sugar source (like light malt extract or honey) with water, sterilize the solution, and then introduce mushroom mycelium. Use a canning jar with an airport lid for the culture and a pressure cooker for sterilization.

Within 7 – 14 days you'll see the mycelium has 'clouded' the culture and expanded throughout; this is aided with semi-regular mixing/shaking. How much liquid culture you need is depending on the medium you are inoculating. If you are using our sterilised grain bags we recommend that you use 10 ml of liquid culture per bag. You can use less but it will slow down the process while increasing the risk of failure.

A good liquid culture will often be a tiny bit cloudy and then clear up. This is because the bacteria starts to grow a tiny bit but the mycelium is so far ahead that it scours the liquid culture of bacteria and particulates and then dominates it (mycelium has been fighting bacteria like this for millions of years).

Light malt extract (LME) recipe: Add 20g of Light Malt Extract (LME) and 2g of yeast extract to 500ml of water and stir until dissolved. This is one of the most commonly used recipes. Karo liquid culture recipe: Add 16g of Karo light corn syrup to 400ml of water and stir until completely dissolved.

Usually about 1 – 2 CC's of liquid culture is sufficient per quart sized grain jar. You can also inoculate the grain jar without even opening the lid, by simply injecting the liquid culture right though the filter on the top of the jar.

To properly incubate your mycelium it must remain at a constant temperature between 70 and 90 degrees depending on what species you are growing. This temperature is also ideal for countless number of other molds and bacteria.

With spore syringes, there's always a risk of contamination, which can ruin your entire batch. Liquid culture eliminates this risk by providing a sterile and controlled environment for the mycelium to grow.

While spore syringes are often filled with a multi-spore solution, liquid culture syringes contain an isolated strain of mushroom mycelium. This difference is crucial for those looking for specific characteristics in their grow.

Inoculate the liquid with a single bacterial colony by touching a sterile inoculating loop to the colony, making certain that some of the cells have been transferred to the loop, and then dipping the loop into the liquid and shaking it a bit. Replace the tube's cap, and place the tube on a roller drum at 60 rpm, 37°C.

Agar is used to identify contaminants, isolate cultures and germinate spores. Clean segments of agar are used to inoculate sterilised grains or broth to produce grain spawn and liquid culture.

The inoculation process is also far shorter in liquid cultures. You can expect mushrooms to only incubate for 10 days to three weeks, which is significantly less than normal. The level of mycelium growth is practically limitless, giving you a high yield every time.
